Abstract: Cancer develops through serial acquisition of somatic genetic alterations and clonal evolution. Clonal hematopoiesis (CH) represents an early stage of blood cancer in which a large fraction of blood cells originates from a single hematopoietic stem cell lineage. CH with cancer driver genetic variants can be identified in healthy individuals with no signs of blood malignancies. CH is rare at young age and its prevalence increase exponentially with age. Presence of CH is associated with increased risk of blood cancer, mortality, and several age-related diseases including, cardiovascular, pulmonary, and liver diseases. Thanks to the large-scale exome and genome sequencing projects that have fueled the studies of CH in the past decade. Recently, we characterized CH with somatic variants in several new driver genes and classified CH into myeloid and lymphoid. These newly defined CH classes enabled the stratification of risk of developing myeloid and lymphoid malignancies. Moving forward, my research group is focused on identification of new drivers of CH and understanding the mechanisms of clonal expansion in blood.
